1. Safety and Preparation

UV lamps can cause eye damage, and lamp covers become very hot during operation, so never look directly at the beam and allow lamps to cool before handling . Always wear safety goggles and gloves, and keep your workspace clean and dry . Check that all warning labels on the device are intact and readable .

2. Instrument Setup

  • Power and Warm-Up: Connect the spectrometer to a stabilized power source and allow the lamps to warm up for at least 15–30 minutes. This ensures thermal stability and prevents baseline drift .
  • Software Initialization: Launch the control software (e.g., ASpect UV for SPECORD PLUS) and clear any hardware flags or communication errors .
  • Light Source Selection: Choose the appropriate lamp for your wavelength range. Use a deuterium lamp for UV and a tungsten-halogen lamp for visible light to optimize energy efficiency and lamp life .

3. Sample Preparation

  • Use clean, clear cuvettes and handle them by the frosted sides to avoid fingerprints, which can alter readings .
  • Rinse cuvettes with deionized water or the sample solution to prevent contamination .
  • Load the correct volume of sample (typically 1 mL for cuvettes) and ensure the light path passes entirely through the liquid .

4. Calibration and Blanking

  • Perform a blank measurement using a cuvette filled with solvent only. This sets the baseline for absorbance measurements .
  • Ensure the optical path is clear and the detector is functioning properly. Calibration may include wavelength verification and photometric checks depending on the instrument .

5. Measurement

  • Insert the sample cuvette into the sample compartment and close the lid.
  • Select the desired wavelength or scan range in the software.
  • Start the measurement. The detector will record the transmitted light and calculate absorbance or transmission values .
  • Observe the spectrum for expected peaks and smooth curves. Peaks correspond to chromophores in the sample absorbing specific wavelengths .

6. Data Interpretation

  • Absorbance values indicate the concentration of the analyte according to Beer-Lambert law.
  • Compare spectra to reference standards or known compounds to identify unknown substances .
